#1 - Spontaneous and experimental poisoning by Myrocarpus frondosus in cattle

Abstract in English:

An outbreak of photosensitization in Holstein and Jersey cattle occurred after the passage of an extratropical cyclone in Southern Brazil. On that occasion, several trees fell, including some of the species Myrocarpus frondosus (Fabaceae family) popularly known as “gabreúva”, “cabriúna” or “gabriúna”. Five heifers consumed the leaves of this plant, and all of them became ill. The main clinical signs were decreased appetite, permanence in the water, decreased ruminal movements, dry stools, congested and icteric sclera and conjunctivae, dark urine and restlessness when exposed to the sun. One heifer showed circling, incoordination, restlessness and death six days after the onset of clinical signs. From the seventh day, three heifers showed improvement; one remained apathetic, with jaundiced mucous membranes, ulceration, and scaling on the tongue, increased respiratory rate, and fever, and died 18 days after ingestion of the plant. At necropsy, photosensitivity lesions were observed on the depigmented skin, snout, teats, and ear tip, characterized by discontinuity of the skin with the formation of crusts, inflammatory exudate associated with redness of the skin, in addition to ulceration in the mouth, snout and ventral portion of the tongue and jaundice and edema in the submandibular region. The liver was enlarged, with an orange color, full gallbladder, and wall edema. Through microscopy, liver lesions were characterized by moderate and diffuse necrosis of hepatocytes, vacuolar and centrilobular degeneration, bilirubin retention and proliferation of biliary epithelium. The literature shows no reports of M. frondosus toxicity in cattle. Experiments were carried out in cattle to clarify the possible clinical-pathological picture produced by this plant. A survey of the epidemiological data of the diagnosed spontaneous outbreak was carried out, and subsequently, three cattle received the plant’s green leaves, and a fourth received the dried leaves. After administering the plant, blood samples were collected daily for blood count, measurement of liver enzymes (gamma glutamyl transferase, alkaline phosphatase, alanine aminotransferase, aspartate transferase), bilirubin, measurement of vital parameters and liver biopsy from the onset of signs. In case of death, a necropsy was performed with collection of viscera samples for macro and microscopic evaluation. The experimental part was developed at the Laboratory of Animal Pathology, CAV-UDESC. The single dose of 33g/kg of green leaves caused severe clinical signs, while the administration of 22g/kg and 11g/kg caused mild to moderate signs of the disease. The supply of desiccated leaves in a single dose did not cause clinical manifestations. M. frondosus can be blamed as a cause of spontaneous hepatogenous photosensitization in cattle. Experimentally, the ingestion of green leaves of this plant, in doses greater than 11g/kg can lead to clinical changes, with subsequent recovery, or progress to death with severe liver damage.

Um surto de fotossensibilização em bovinos da raça Holandesa e Jersey foi observado após a passagem de um ciclone extratropical no Sul do Brasil, quando houve queda de diversas árvores, dentre elas Myrocarpus frondosus (família Fabaceae), popularmente conhecida como “gabreúva, cabriúna ou gabriúna”. Cinco novilhas consumiram folhas dessa planta, e todas adoeceram. Os principais sinais clínicos foram diminuição do apetite, permanência dentro da água, diminuição dos movimentos ruminais, fezes ressecadas, esclera e conjuntivas congestas e ictéricas, urina escura e inquietação quando expostas ao sol. Uma novilha apresentou andar em círculos, incoordenação e inquietação e morte seis dias após início dos sinais clínicos. A partir do sétimo dia, três novilhas apresentaram melhora e uma permaneceu apática, com as mucosas ictéricas, ulceração e descamação na língua, aumento da frequência respiratória, febre e morreu 18 dias após ingestão da planta. Na necropsia foram observadas lesões de fotossensibilização na pele despigmentada, focinho, tetos, ponta da orelha, caracterizadas por descontinuidade da pele com formação de crostas, exsudato inflamatório associado a vermelhidão da pele além de ulceração na boca, focinho e porção ventral da língua e icterícia acentuada e edema, na região submandibular. O fígado estava aumentado de volume, de cor laranja, vesícula biliar repleta e com edema de parede. Através da microscopia foram observadas, lesões hepáticas, caracterizadas por necrose de hepatócitos, moderada e difusa, degeneração vacuolar, centrolobular, retenção de bilirrubina e proliferação de epitélio biliar. Na literatura, não há relatos da toxicidade de M. frondosus para bovinos. Com o objetivo de esclarecer o possível quadro clínico-patológico produzido por essa planta foram conduzidos experimentos em bovinos. Foi realizado um levantamento dos dados epidemiológicos do surto espontâneo diagnosticado e, posteriormente, três bovinos receberam as folhas verdes da planta e um quarto recebeu as folhas dessecadas. Após a administração da planta, foram coletadas amostras de sangue diariamente para hemograma, dosagem de enzimas hepáticas (gama glutamil transferase, fosfatase alcalina, alanina aminotransferase, aspartato transferase), bilirrubina, aferição dos parâmetros vitais e biópsia hepática a partir do surgimento dos sinais clínicos. Em caso de morte, foi realizada necropsia com coleta de amostras de vísceras para avaliação macro e microscópica. A parte experimental foi desenvolvida nas dependências do Laboratório de Patologia Animal, CAV-UDESC. A dose única de 33g/kg de folhas verdes causou graves sinais clínicos, enquanto a administração de 22g/kg e 11g/kg causou sinais leves a moderados da doença. O fornecimento de folhas dessecadas em dose única não causou manifestações clínicas. M. frondosus pode ser responsabilizada como causa de fotossensibilização hepatógena espontânea em bovinos. A ingestão de folhas verdes dessa planta em doses superiores a 11g/kg pode levar a alterações clínicas, com posterior recuperação, ou evoluir para morte com lesões hepáticas graves.


#2 - Dorsal thermal necrosis in a Brazilian sheep flock

Abstract in English:

Thermal burn injuries are very uncommon occurrences in small ruminants worldwide, and in Brazil, there are no descriptions of this condition. Herein we describe an outbreak of dorsal thermal necrosis in a flock of sheep in the municipality of Boca da Mata, in the Forest Zone of the State of Alagoas, Northeastern Brazil. Twelve ewes were separated from the flock for a general clinical examination and evaluation the serum levels of urea, creatinine, gamma-glutamyltransferase, and aspartate aminotransferase. In addition, biopsies of the skin and liver were performed on two severely affected sheep. The flock was raised in semi-intensively management in a pasture area without shading and receiving approximately eight hours of insolation daily and developed severe dermatitis that evolved to severe necrosis of the dorsal skin. Grossly, these lesions were only observed in the dorsal skin of black sheep, while sheep with white dorsal areas of skin were not affected. Microscopically, the epidermis and deep dermis of affected sheep showed diffuse coagulative necrosis, pigmentary incontinence, and parakeratotic hyperkeratosis. Dorsal thermal necrosis occurs only in the dorsal skin of black sheep in situations of excessive exposure to sunlight due to the accumulation of ultraviolet radiation. The distribution of the lesions, restricted to the dorsal area of the skin, is an important clinicopathological aspect in these cases that can help differentiate lesions caused by photosensitization, as they usually involve other anatomical sites. To the authors’ knowledge, this is the first report of dorsal thermal necrosis in sheep in Brazil.

RESUMO.- [Necrose térmica dorsal em rebanho ovino brasileiro.] Lesões por queimaduras térmicas são ocorrências muito incomuns em pequenos ruminantes no mundo e no Brasil não há registros da ocorrência desta condição. Descreve-se um surto de necrose térmica dorsal em um rebanho de ovinos no município de Boca da Mata, Zona da Mata do Estado de Alagoas, Nordeste do Brasil. Doze ovelhas foram separadas do rebanho para realização de exame clínico geral e avaliação dos níveis séricos de uréia, creatinina,gama-glutamiltransferase e aspartato aminotransferase. Adicionalmente, biópsias de pele e fígado foram realizadas em duas ovelhas severamente afetadas. O rebanho era criado sob manejo semi-intensivo em uma área de pastagem sem sombreamento e recebiam aproximadamente oito horas de insolação diárias. Essas ovelhas desenvolveram dermatite severa que evoluiu para necrose da pele dorsal. Macroscopicamente, as lesões foram observadas apenas na pele dorsal de ovelhas negras, enquanto ovelhas com áreas dorsais brancas da pele não foram afetadas. Microscopicamente, a epiderme e a derme profunda das ovelhas afetadas apresentavam necrose coagulativa difusa, além de incontinência pigmentar e hiperqueratose paraqueratótica. A necrose térmica dorsal ocorre apenas na pele dorsal de ovelhas negras em situações de exposição excessiva à luz solar devido ao acúmulo de radiação ultravioleta. A distribuição das lesões, restrita à região dorsal da pele, é um importante aspecto clínico-patológico nesses casos que pode auxiliar na diferenciação das lesões causadas por fotossensibilização, pois geralmente envolvem outros sítios anatômicos. Este é o primeiro relato de necrose térmica dorsal em ovinos no Brasil.


#3 - Bilateral perirenal fat necrosis in a sambar deer (Rusa unicolor)

Abstract in English:

The gross and histopathological findings of perirenal fat necrosis in a sambar deer (Rusa unicolor) are described in a 6-year-old female deer that was necropsied after showing severe apathy, weight loss, recumbency, and death. At post-mortem examination, two large white to yellow, irregular, and encapsulated masses involving both kidneys were observed. Microscopically, these masses consisted of fat tissue presenting multifocal to coalescing areas of necrosis, diffuse infiltration of lymphocytes, macrophages, occasional plasma cells, and multinucleated giant cells. Multifocally, the necrotic sites were replaced by mineralization and fibrosis. The described condition has not been reported in deer and the exact mechanisms of development of perirenal fat necrosis remain to be determined. Massive fat necrosis has been reported in other animal species such as dogs, cats, pigs, goats, cattle, and whales (Balaena mysticetus). It is usually an incidental finding during post-mortem examination that needs to be differentiated from neoplasms originating from the adipose tissue, kidneys and adrenals, and disorders resulting from pancreatitis and obesity.

São descritos os achados anátomo e histopatológicos da necrose gordurosa perirrenal em um cervo sambar (Rusa unicolor) fêmea de seis anos de idade que foi necropsiado após apresentar severa apatia, perda de peso, decúbito permanente e morte. Durante a necropsia foram observadas duas grandes massas brancas a amarelas, duras, irregulares e encapsuladas envolvendo ambos os rins. Microscopicamente, essas massas consistiam em áreas de necrose multifocais a coalescentes do tecido adiposo perirrenal, com infiltrado difuso de linfócitos, macrófagos e ocasionais plasmócitos e células gigantes multinucleadas. Multifocalmente, as áreas necróticas estavam substituídas por mineralização e fibrose. A compressão dessas massas no córtex renal foi associada à degeneração tubular renal leve a mínima. A condição descrita não foi relatada em veados e os mecanismos exatos de desenvolvimento da necrose da gordura perirrenal ainda precisam ser determinados. Necrose gordurosa maciça foi relatada em outras espécies de animais, como caninos, felinos, suínos, caprinos, bovinos e baleias (Balaena mysticetus). Geralmente é um achado incidental durante o exame post-mortem que precisa ser diferenciado de neoplasias originárias do tecido adiposo, rins e adrenais, e distúrbios decorrentes de pancreatite e obesidade.


#4 - Acute necrotic hepatotoxicity caused by Lantana camara L. ingestion in dairy cattle

Abstract in English:

This study describes an outbreak of acute necrotic hepatopathy associated with spontaneous poisoning by Lantana camara L. in dairy cattle. A herd of 15 cows and heifers was introduced into a native pasture with limited food supply, and, sixteen days later, eight animals had spontaneous nasal hemorrhage, fever, lethargy, jaundice, and dry, dark stools with mucus and blood. The clinical course varied from two to five days. In the pasture where the cattle were kept, abundant adult specimens of L. camara L. with evident signs of consumption were observed. In total, seven cattle died and necropsy was performed in three of them. All animals had moderate jaundice, hemorrhage in the subcutaneous tissue and on the surface of different organs. The liver was slightly enlarged, with orange discoloration and enhanced lobular pattern. Histologically, multifocal areas of coagulative necrosis of hepatocytes in the centrilobular area, occasionally extending to the midzonal area, were observed, as well as marked hepatocellular degeneration and prominent cholestasis. The current study suggests that L. camara L. poisoning should be considered a differential diagnosis of acute and necrotic hepatotoxicity in cattle, despite the absence of photosensitization.

Esse estudo descreve um surto de hepatopatia necrótica aguda associada a intoxicação espontânea por Lantana camara L. em bovinos leiteiros. Um lote de 15 vacas e novilhas foram introduzidas em um piquete com campo nativo, com escassa oferta de alimento. Após dezesseis dias, oito animais manifestaram epistaxe, febre, apatia, icterícia, fezes ressecadas e escuras com muco e sangue. A evolução do quadro clínico variou de dois a cinco dias. No piquete em que os bovinos estavam alojados havia grande quantidade de L. camara L. com sinais evidentes de consumo. No total, sete bovinos morreram, e destes, o exame post mortem foi realizado em três. Os bovinos exibiam moderada icterícia, hemorragias no tecido subcutâneo e na superfície de diferentes órgãos. O fígado estava discretamente aumentado, com coloração alaranjada e padrão lobular evidente. As lesões histológicas consistiam em acentuada necrose de coagulação de hepatócitos em região centrolobular, por vezes se estendendo a região mediozonal, além de acentuada degeneração dos hepatócitos e evidente Colestase. O presente trabalho alerta para que intoxicação por L. camara L. seja levada em consideração nos diagnósticos diferenciais de hepatotoxicidade necrótica aguda em bovinos, mesmo sem indícios de fotossensibilização.


#5 - Spontaneous and experimental poisoning by nitroxinil at 34% in goats

Abstract in English:

This study describes the epidemiological, clinical, and pathological aspects of spontaneous and experimental poisoning by nitroxinil at 34% concentration in goats. The outbreak occurred on a farm in the municipality of Prata, Paraíba state. Nitroxinil was administered to a herd of 120 goats, of which 18 presented with anorexia, vocalization, abdominal distension, weakness, staggering, and falls. Necropsy of three goats revealed that the main lesion was acute liver injury. Histologically the liver showed centrilobular necrosis associated with hemorrhage and hepatocyte degeneration. In the kidneys, tubular nephrosis with granular cylinder formations was observed. The lungs showed multifocal to coalescent areas of moderate interalveolar edema and vascular congestion. Experimental poisoning was carried out in two goats, with the same medication and doses administered on the farm. The experimental goats showed clinical signs and macroscopic and histological changes similar to the spontaneously poisoned goats. The diagnosis of nitroxinil poisoning was made based on epidemiological, clinical, and pathological data, and confirmed by experimental poisoning. The administration of nitroxinil in high doses, associated with high ambient temperature and physical exercises, can cause poisoning with high lethality in goats.

Este estudo descreve os aspectos epidemiológicos, clínicos e patológicos da intoxicação espontânea e experimental por nitroxinil na concentração de 34% em caprinos. O surto ocorreu em uma fazenda no município de Prata, Paraíba. Nitroxinil foi administrado a um rebanho de 120 cabras, das quais 18 apresentavam anorexia, vocalização, distensão abdominal, fraqueza, cambaleando e quedas. A necropsia de três cabras revelou que a lesão principal era uma lesão hepática aguda. Histologicamente, o fígado apresentava necrose centrolobular associada a hemorragia e degeneração de hepatócitos. Nos rins, nefrose tubular com formações de cilindro granular foi observada. Os pulmões apresentavam áreas multifocais a coalescentes de edema interalveolar moderado e congestão vascular. A intoxicação experimental foi realizada em duas cabras, com a mesma medicação e doses administradas na fazenda. As cabras experimentais apresentaram sinais clínicos e alterações macroscópicas e histológicas semelhantes às cabras intoxicadas espontaneamente. O diagnóstico de intoxicação por nitroxinil foi feito com base em dados epidemiológicos, clínicos e patológicos, e confirmado por intoxicação experimental. A administração de nitroxinil em altas doses, associada à alta temperatura ambiente e exercícios físicos, pode causar intoxicação com alta letalidade em caprinos.


#6 - Spontaneous poisoning by Dodonaea viscosa (Sapindaceae) in cattle in Southern Brazil

Abstract in English:

In this study, an outbreak of spontaneous poisoning by Dodonaea viscosa (D. viscosa) in a herd of dairy cattle in the municipality of Capão do Leão, Rio Grande do Sul, was investigated. Three deaths occurred in a batch of 16 Jersey cattle, aged between three and four years, kept in a native field. The clinical signs observed were apathy, decreased production, and anorexia, with death occurring within approximately 48 h after the onset of signs. The three cattle were necropsied, and tissue samples were sent for histopathological examination. Necropsy findings included serosanguineous fluid in the abdominal cavity, intestines with congested serosa, and marked mesenteric edema. The mucosa of the abomasum of two of the animals was hemorrhagic with bloody content, and among the ruminal content of a bovine, leaves with morphological characteristics compatible with D. viscosa were observed. The livers of the three animals were enlarged, with accentuation of the lobular pattern. Histologically, centrilobular coagulation necrosis with congestion and hemorrhage was observed in the liver. Vacuolization and degeneration of hepatocytes were observed in the mid-zonal and periportal regions. The diagnosis of poisoning by D. viscosa leaves was based on epidemiological data, necropsy findings, and histopathological alterations. The presence of the plant in the rumen and in the grazing site of the affected cattle was essential for the diagnosis.

Neste trabalho, é descrito um surto de intoxicação espontânea por Dodonaea viscosa (D. viscosa) ocorrido em um rebanho de bovinos leiteiros, no município de Capão do Leão, no Rio Grande do Sul. Ocorreram três mortes em um lote de 16 bovinos da raça Jersey com idades entre três e quatro anos, mantidos em campo nativo. Os sinais clínicos observados foram apatia, queda na produção e anorexia, com morte em aproximadamente 48 horas após o início dos sinais. Os três bovinos foram necropsiados, e amostras de tecidos foram encaminhadas para exame histopatológico. Os achados de necropsia incluíam líquido serossanguinolento na cavidade abdominal, intestinos com serosas congestas e marcado edema de mesentério. A mucosa do abomaso de dois animais apresentava-se hemorrágica com conteúdo sanguinolento e, em meio ao conteúdo ruminal de um bovino foram observadas folhas com caracteres morfológicos compatíveis com D. viscosa. O fígado dos três animais estava aumentado, com acentuação do padrão lobular. Histologicamente no fígado havia necrose de coagulação centrolobular com congestão e hemorragia. Nas regiões médio-zonal e periportal observou-se vacuolização e degeneração dos hepatócitos. O diagnóstico de intoxicação pelas folhas D. viscosa foi baseado nos dados epidemiológicos, nos achados de necropsia e nas alterações histopatológicas. A presença da planta no rúmen e no local de pastoreio dos bovinos afetados foi fundamental para o diagnóstico.


#7 - Clostridium perfringens α and β recombinant toxoids in equine immunization

Abstract in English:

Clostridium perfringens is considered one of the main causative agents of superacute enterocolitis, usually fatal in the equine species, due to the action of the β toxin, and is responsible for causing severe myonecrosis, by the action of the α toxin. The great importance of this agent in the equine economy is due to high mortality and lack of vaccines, which are the main form of prevention, which guarantee the immunization of this animal species. The aim of this study was to evaluate three different concentrations (100, 200 and 400µg) of C. perfringens α and β recombinant toxoids in equine immunization and to compare with a group vaccinated with a commercial toxoid. The commercial vaccine was not able to stimulate an immune response and the recombinant vaccine was able to induce satisfactory humoral immune response in vaccinated horses, proving to be an alternative prophylactic for C. perfringens infection.

Clostridium perfringens é considerado um dos principais agentes causadores de enterocolites superagudas, geralmente fatais na espécie equina, devido à ação da toxina β, além de ser responsável por causar quadros graves de mionecrose, pela ação da toxina α. A grande importância desses agentes na equinocultura, deve-se a elevada mortalidade e a inexistência de vacinas, principal forma de prevenção, que garantam a imunização dessa espécie animal. O objetivo deste trabalho foi avaliar três diferentes concentrações (100, 200 e 400µg) dos toxóides recombinantes α e β de C. perfringens na imunização de equinos, bem como comparar com um grupo vacinado com um toxóide comercial. A vacina comercial não se mostrou capaz de estimular uma resposta imune e a vacina recombinante foi capaz de induzir resposta imune humoral satisfatória em equinos vacinados, provando ser uma alternativa profilática para infecção por C. Perfringens.

Serum activities of creatine kinase were increased in all sheep examined. Multifocal polyphasic muscle segmental degeneration and necrosis was observed in skeletal muscles. Although the plant is a common weed in northeastern Brazil, the poisoning is rare, probably because the animals do not ingest it or due to toxicity variations.

As atividades séricas da creatina quinase foram aumentadas em todos os ovinos examinados. Foi observada degeneração segmentar e necrose polifásica muscular em músculos esqueléticos. Embora a planta seja uma erva daninha comum no Nordeste do Brasil, o envenenamento é raro, provavelmente porque os animais não o ingerem ou devido a variações de toxicidade.

E-mail: dantas.af@pq.cnpq.br We described the main epidemiological, clinical and pathological aspects of canine infectious hepatitis diagnosed in the Laboratory of Animal Pathology of the Federal University of Campina Grande during the period from January 2003 to December 2016. Of the 1,640 necropsied dogs, 15 were diagnosed as infectious canine hepatitis (0.91%). Of the dogs affected nine were males and six females. The ages ranged from 45 days to seven years, being most of them young. Ten animals were mixed breed, four were Poodles and one Rottweiler. Most of the dogs do not received any vaccine protocol. The dogs came from the municipalities of Patos, São Mamede and Teixeira, from Paraiba, northeastern of Brazil. Most of the dogs presented clinical course varying from hyperacute to acute. The main clinical signs were seizure, apathy and hyporexia. At necropsy, the major alterations were observed in the liver, which was pale to orange and with irregular reddish areas on the capsular surface, besides accentuation of the lobular pattern and edema in the wall of the gallbladder. Hemorrhages were observed in several organs. In the histopathology there was centrolobular necrosis of hepatocytes associated with intranuclear viral inclusion bodies, hemorrhages and mixed inflammatory infiltrate. The diagnosis was established based on the characteristic histopathological lesions and was confirmed by immunohistochemistry. Infectious canine hepatitis occurs occasionally in the Paraiba, affecting particularly young and unvaccinated dogs.

A maioria dos cães não recebeu nenhum tipo de protocolo vacinal. Os cães eram oriundos dos municípios de Patos, São Mamede e Teixeira, pertencentes ao estado da Paraíba, Nordeste do Brasil. A maioria dos cães apresentou curso clínico variando de hiperagudo a agudo. Os principais sinais clínicos foram convulsão, apatia e hiporexia. Na necropsia, as principais alterações foram observadas no fígado que se apresentava de pálido a alaranjado e com áreas irregulares avermelhadas na superfície capsular, além de acentuação do padrão lobular e edema na parede da vesícula biliar. Hemorragias foram observadas em vários órgãos. Na histopatologia havia necrose centrolobular de hepatócitos associada a corpúsculos de inclusão viral intranucleares, hemorragia e infiltrado inflamatório misto. O diagnóstico foi estabelecido com base nas características lesões histopatológicas e foi confirmado por imuno-histoquímica. A hepatite infecciosa canina ocorre ocasionalmente na Paraíba, acometendo particularmente cães jovens e não vacinados.


#10 - Comparison of the effects of Cestrum axillare leaves with isolated saponins in goats

Abstract in English:

Cestrum axillare Vell. (formerly Cestrum laevigatum Schltd.), family Solanaceae, is the most important hepatotoxic plant in Brazil that causes acute poisoning. It occurs in the Southeast and Center-West regions and in coastal areas of the Northeast Brazil. Spontaneous poisoning was described in cattle, goats and sheep, with clinical signs evidenced within 24 hours after ingestion of the leaves and death within 48 hours after signs onset. The clinical signs observed in acute poisoning are apathy, anorexia, ruminal arrest, arched back, constipation with feces in small spheres, sometimes covered with mucus and blood streaks, muscle tremors, staggering gait and sometimes sialorrhoea. Neurological signs may be observed, due to interference in the urea cycle due to hepatic insufficiency resulting in hyperammonemia (hepatic encephalopathy). The main pathological finding is centrilobular hepatic necrosis. The toxic principle present in C. axillare was not yet definitively proven, but some authors attribute the toxicity of the plant to the presence of saponins gitogenin and digitogenin. However, it has not been determined whether the saponins present in C. axillare are responsible for the hepatotoxic effect of the plant. Thus, the objective of this work is to determine if the saponins are the compounds responsible for the hepatotoxic effects produced by the ingestion of the leaves of C axillare, using goats as experimental model. For this, the effects of the administration of the leaves were compared with those produced by the saponins isolated from the leaves in goats. Six goats were randomly assigned to three experimental groups that received [1] dry leaves of C. axillare (animals A1 and A2), [2] saponins extract from leaves (animals S1 and S2) or [3] control group (animals C1 and C2). For goats receiving the dry leaves the administered dose of plant was 10g/kg for one animal (A1) and 5g/kg for the other one (A2). For animals receiving the saponins extract, administration was done at a dose equivalent to 20g/kg repeated after 24 hours. The dry leaves administered at a dose of 10g/kg to a goat produced toxic effects, with alterations in biochemistry (indicating hepatic lesion) and histopathology showing centrilobular hepatic necrosis. At the dose of 5 g/kg of dry leaves, clinical signs of poisoning were not observed, but hepatic necrosis was found; after 15 days after the last administration, the hepatic parenchyma of this animal was already normal, with only hemorrhagic areas, demonstrating full regeneration. The administration of extracts of saponins containing gitogenin and digitogenin to goats did not produce significant toxic effects, proving that these compounds are not responsible for intoxication. In addition, goats are a good experimental model for studies of this intoxication.

Cestrum axillare Vell. (anteriormente C. laevigatum Schltd.), família Solanaceae, é a mais importante planta hepatotóxica do Brasil que causa intoxicação aguda. Tem ocorrência nas regiões Sudeste e Centro-Oeste e em áreas litorâneas do Nordeste. A intoxicação natural foi descrita em bovinos, caprinos e ovinos, com sinais clínicos evidenciados em até 24 horas após a ingestão das folhas e morte em até 48 horas após o início da sintomatologia. Os sinais clínicos observados na intoxicação aguda são apatia, anorexia, parada ruminal, dorso arqueado, constipação com fezes em formas de pequenas esferas, por vezes recobertas com muco e com estrias de sangue, tremores musculares, andar cambaleante e, às vezes, sialorreia. Podem ser observados sinais neurológicos, devido à interferência no ciclo da ureia pela insuficiência hepática resultando em hiperamonemia (encefalopatia hepática). O principal achado patológico é a necrose hepática centrolobular. O princípio tóxico presente no C. axillare ainda não está definitivamente comprovado, mas alguns autores atribuem a toxicidade da planta à presença das saponinas gitogenina e digitogenina. No entanto, ainda não foi determinado se as saponinas presentes em C. axillare são as responsáveis pelo efeito hepatotóxico da planta. Assim, o objetivo deste trabalho é determinar se as saponinas são os compostos responsáveis pelos efeitos hepatotóxicos produzidos pela ingestão das folhas de C. axillare, usando caprinos como modelo experimental. Para isto, foram comparados os efeitos da administração das folhas com os produzidos pelas saponinas isoladas destas folhas em caprinos. Foram utilizados seis caprinos, distribuídos aleatoriamente em três grupos experimentais que receberam [1] folhas secas de C. axillare (Caprinos A1 e A2), [2] extrato de saponinas das folhas (Caprinos S1 e S2), e [3] grupo controle (Caprinos C1 e C2). Para os caprinos que receberam as folhas secas a dose administrada de planta foi de 10g/kg para um animal (A1) e de 5g/kg para outro (A2). Para os animais que receberam o extrato de saponinas, a administração foi feita na dose equivalente a 20g/kg, repetida após 24 horas. Foi verificado que as folhas secas, quando administradas na dose de 10g/kg a um caprino, produziram efeitos tóxicos, com alterações na bioquímica (indicando lesão hepática) e histopatológica apresentando necrose hepática centrolobular. Na dose de 5g/kg de folhas secas, não foi observado sintomatologia clínica da intoxicação, mas houve necrose hepática; 15 dias após a última administração, o parênquima hepático deste animal já se encontrava normal, apenas com áreas hemorrágicas, demonstrando plena regeneração. A administração do extrato de saponinas contendo gitogenina e digitogenina a caprinos não produziu efeitos tóxicos significantes, comprovando não serem estes compostos os responsáveis pela intoxicação. Além disto, a espécie caprina é um bom modelo experimental para estudos desta intoxicação.
