#21 - Evaluation of five parasitological techniques for diagnosing phylum Ciliophora cysts in fecal samples from free-living wild artiodactyls

Abstract in English:

A variety of laboratory techniques are used in parasitological diagnosis. However, studies that analyze their laboratory efficiency are very scarce, especially with regard to biological samples from wild animals that are little known, with little popular attachment, such as artiodactyls. These can be infected by different parasites, including protozoa of the phylum Ciliophora, which includes the parasites Balantioides coli and Buxtonella sulcata. In this light, the aim of this study was to compare the efficiency of five coproparasitological techniques for diagnosing protozoan cysts of the phylum Ciliophora in the feces of free-living artiodactyls. To this end, 101 fecal samples were collected from trails in Pedra Selada State Park, Rio de Janeiro state, from 2020 to 2021. All the samples were analyzed using the qualitative techniques of modified Sheather floatation, modified Ritchie sedimentation and Lutz, as well as the quantitative techniques of Mini-FLOTAC and McMaster. Cyst recovery was best achieved using the modified Ritchie technique, in which 62.5% positivity was detected, followed by Lutz (47.5%), modified Sheather (37.5%) and the quantitative techniques of Mini-FLOTAC (30%) and McMaster (17.5%). In most of the comparisons between the techniques, reasonable agreement regarding the diagnosis was observed (Kappa 0.21 to 0.40), which was statistically significant (p≤0.05). McMaster showed higher mean and standard deviation values for counts of cysts per gram of feces than Mini-FLOTAC. However, there was no significant difference in the estimates for cyst counts (Wilcoxon p>0.05). Sedimentation qualitative techniques were more indicated for diagnosing cysts of protozoa of the phylum Ciliophora in the feces of free-living wild artiodactyls. These techniques can therefore be used as laboratory tools for environmental parasite monitoring. In addition, between the two quantitative techniques, Mini-FLOTAC presented better performance, thus showing its potential as a tool for estimating the abundance of cystic forms of the phylum Ciliophora in environmental samples.

Diversas são as técnicas laboratoriais utilizadas no diagnóstico parasitológico. No entanto, estudos que analisam a eficiência laboratorial das mesmas são muito escassos, principalmente quando se trata de amostras biológicas de animais silvestres, que possuem pouca divulgação e apego popular, como os artiodáctilos. Estes podem se infectar por diferentes parasitos, incluindo os protozoários do Filo Ciliophora, no qual se inclui Balantioides coli, bem como Buxtonella sulcata. Mediante o exposto, este estudo objetivou comparar a eficiência entre cinco técnicas coproparasitológicas para o diagnóstico de cistos de protozoário do Filo Ciliophora em fezes de artiodáctilos em vida livre. Entre 2020 e 2021 foram coletadas 101 amostras fecais em trilhas do Parque Estadual da Pedra Selada localizado no estado do Rio de Janeiro. Todas as amostras foram analisadas pelas técnicas qualitativas de flutuação de Sheather modificada, sedimentação de Ritchie modificada e Lutz, bem como por meio das técnicas quantitativas de Mini-FLOTAC e McMaster. Pode-se verificar uma superioridade na recuperação dos cistos por meio da técnica de Ritchie modificada, no qual foi detectado 62,5% de positividade, seguida pelo Lutz (47,5%), Sheather modificada (37,5%) e pelas técnicas quantitativas de Mini-FLOTAC (30%) e McMaster (17,5%). Na maioria das comparações entre as técnicas foi possível verificar uma concordância razoável (Kappa 0,21 a 0,40) em relação ao diagnóstico entre as mesmas, sendo este estatisticamente significativo (p≤0,05). McMaster em comparação com o Mini-FLOTAC foi a que apresentou os maiores valores médio e de desvio padrão na contagem dos cistos por grama de fezes. No entanto, não foi evidenciado diferença significativa na estimativa de contagem dos cistos (Wilcoxon=p>0,05). Pode-se verificar que as técnicas qualitativas de sedimentação foram consideradas as mais indicadas para o diagnóstico de cistos do protozoário do Filo Ciliophora em fezes de artiodáctilos silvestres em vida livre, podendo ser utilizadas como ferramentas laboratoriais de monitoramento parasitário ambiental. Além disso, dentre as técnicas quantitativas, o Mini-FLOTAC foi a que apresentou a melhor performance, denotando o seu potencial como uma ferramenta para se estimar a abundância de formas císticas do Filo Ciliophora em amostra ambiental.


#22 - Occurrence of Salmonella spp. in fecal samples from foals with and without diarrhea in the state of São Paulo: microbiological diagnosis, antimicrobial susceptibility profile, and molecular detection

Abstract in English:

The present study investigated Salmonella spp. in the feces of 200 foals up to one year of age (100 with clinical signs of diarrhea and 100 without clinical signs of diarrhea). Bacteriological culture, serotyping, antimicrobial susceptibility, and real-time PCR (qPCR SYBR® Green or a TaqMan®) for detecting the invA gene (with and without a selective pre-enrichment step in tetrathionate broth) were performed. Bacterial culture revealed 15% (n=30) of positive animals (21 animals with diarrhea and nine without diarrhea). Among the 30 isolates, 13 different serovars were identified: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multidrug resistance was found in 43.33% (n=13) of the isolates, with one isolate obtained from animals without diarrhea and 12 isolates from animals with diarrhea. All qPCR techniques used in the study classified more samples as positive for Salmonella spp. than the bacterial culture of feces. In addition, all qPCR techniques detected more positive animals in the diarrhea group than in the diarrhea-free group. The results confirm the utility of the qPCR method without the pre-enrichment step in tetrathionate as a rapid test for Salmonella spp. in carrier animals. In animals with clinical signs of diarrhea, it can be combined with bacterial culture (antimicrobial susceptibility testing and serotyping). The isolation of Salmonella spp. in nine animals without diarrhea confirms the importance of asymptomatic carrier animals in the epidemiology of the disease. The multidrug resistance observed highlights the importance of rational antimicrobial use in horses and adopting biosecurity protocols that are efficacious in controlling the spread of infections between animals and zoonotic transmission in farms.

O presente estudo investigou a ocorrência de Salmonella spp. em fezes de 200 potros com até um ano de idade (100 com sinais clínicos de diarreia e 100 sem sinais clínicos de diarreia), utilizando as técnicas de cultivo bacteriológico e PCR em tempo real (qPCR) pelos métodos de corante fluorescente (SYBR® Green) e sonda específica (Taqman®) para a detecção do gene invA com e sem etapa de pré-enriquecimento seletivo em caldo de tetrationato. O cultivo bacteriológico revelou 15% (n=30) de animais positivos (21 animais com diarreia e nove animais sem diarreia). Dentre esses 30 isolados, 13 sorovares diferentes foram identificados: S. Infantis, S. Minnesota, S. I.4,5,12:i:-; S. Anatum, S. Cerro, S. Oranienburg, S. Braenderup, S. Give, S. Newport, S. IIIb 61:c:z35, S. 109:-:1.5, S. I.4.12:d:-, S. I.6.8:-:-. Multirresistência foi constatada em 43,33% (n=13) dos isolados, sendo um isolado obtido de animal sem diarreia e 12 isolados de animais com diarreia. Todas as técnicas de qPCR empregadas no estudo apresentaram maior número de amostras classificadas como positivas para Salmonella spp. comparadas ao cultivo bacteriológico de fezes. Adicionalmente, em todas as técnicas de qPCR houve maior número de animais detectados como positivos no grupo de animais com diarreia em relação aos animais sem diarreia. Os resultados confirmaram a utilidade do método qPCR sem a etapa de pré-enriquecimento em tetrationato, como um teste rápido para detecção de Salmonella spp. em animais portadores ou em animais com sinais clínicos de diarreia. O cultivo bacteriológico deve ser associado para a realização do teste de sensibilidade aos antimicrobianos e sorotipificação. O isolamento de Salmonella spp. em nove animais sem diarreia, confirma a importância dos animais portadores assintomáticos na epidemiologia da doença. A multirresistência observada evidencia a importância do uso racional de antimicrobianos em equinos e a importância da adoção de protocolos de biossegurança que sejam eficazes para controlar a disseminação de infecções entre animais e a transmissão zoonótica nas fazendas.


#23 - Real-time PCR quantification and histopathological findings of Mycoplasma hyopneumoniae infection in the lungs of pigs slaughtered in São Luís, Maranhão, Brazil

Abstract in English:

Porcine enzootic pneumonia (PES), mainly caused by the bacteria Mycoplasma hyopneumoniae, is the main cause of respiratory problems in pigs. Infection by M. hyopneumoniae leads to production losses and the predisposition of affected animals to secondary infections, which may result in the condemnation of carcasses and organs due to lung lesions at the time of slaughter. The objective of the research was to evaluate the infection by M. hyopneumoniae in pigs submitted to slaughter in São Luís Island/MA, using molecular and histopathological diagnostic methods. One hundred fifty lung samples were collected from inspected (n=65) and non-inspected (n=85) slaughter pigs on São Luís Island, Maranhão, from July 2019 to August 2021. Of the 150 DNA samples collected, 121 showed an amplified product for Cyt B in the PCR assay. Thus, 121 samples were submitted to qPCR of M. hyopneumoniae, of which 44 (36.36%) showed positive results. The mean amount of bacterial load ranged from 1.20 × 101 to 7.20 × 104, with a mean of 1.73 × 104 copies. Of the reagent samples, 81.81% (36 samples) were obtained from non-inspected slaughter, while 18.18% (8 samples) were obtained from slaughterhouses. In the histopathological analysis, 44 positive qPCR samples were evaluated, of which 28 (63.63%) presented results compatible with the main inflammatory process associated with the presence of M. hyopneumoniae, that is, bronchial-associated lymphoid tissue hyperplasia (BALT). Three samples that showed the highest bacterial load (qPCR: 5.63 × 10³, 2.19 × 104 and 7.23 × 104) showed more evident lesions in this study. The microscopic findings associated with the quantifications indicated a relationship between the amount of bacterial load and the presence of microscopic lesions; higher bacterial load in lung tissue is associated with increased histopathologic staining for BALT hyperplasia. In conclusion, the results point to the circulation of the etiological agent in the sampled animals and the need for preventive measures on pig farms in Maranhão with the involvement of producers, sanitary defense and inspection agencies.

A pneumonia enzoótica suína (PES), causada principalmente pela bactéria Mycoplasma hyopneumoniae, é a principal causa de problemas respiratórios em suínos. A infecção por M. hyopneumoniae leva a perdas produtivas e a predisposição dos animais acometidos a infecções secundárias, o que pode resultar em condenação de carcaças e órgãos por lesões pulmonares no momento do abate. O objetivo da pesquisa foi avaliar a infecção por M. hyopneumoniae em suínos submetidos ao abate na Ilha de São Luís, por meio de métodos diagnósticos moleculares e histopatológicos. Para isso, foram coletadas 150 amostras de pulmão de suínos de abate inspecionado (n=65) e não inspecionado (n=85) na Ilha de São Luís/Maranhão, no período de julho de 2019 a agosto de 2021. Das 150 amostras de DNA coletadas, 121 apresentaram produto amplificado para Cyt B no ensaio de PCR. Assim, 121 amostras foram submetidas à qPCR de M. hyopneumoniae, das quais 44 (36,36%) apresentaram resultados positivos. A quantidade média de carga bacteriana variou de 1,20 × 101 a 7,20 × 104, com média de 1,73 × 104 cópias. Das amostras reagentes, 81,81% (36 amostras) foram obtidas de abate não inspecionado, enquanto 18,18% (8 amostras) foram obtidas em abatedouro. Na análise histopatológica, foram avaliadas 44 amostras positivas para qPCR, das quais 28 (63,63%) apresentaram resultados compatíveis com o principal processo inflamatório associado à presença de M. hyopneumoniae, ou seja, hiperplasia do tecido linfóide associado ao brônquio (BALT). Três amostras que apresentaram maior carga bacteriana (qPCR: 5,63 × 10³, 2,19 × 104 e 7,23 × 104) foram mais evidentes neste estudo. Os achados microscópicos associados às quantificações indicaram uma relação entre a quantidade de carga bacteriana e a presença de lesão microscópica; a maior carga bacteriana no tecido pulmonar está associada a maior alteração histopatológica para hiperplasia BALT. Em conclusão, os resultados obtidos sinalizam para a circulação do agente etiológico nos animais amostrados e a necessidade de medidas preventivas nas criações de suínos do estado do Maranhão com envolvimento dos produtores, órgãos de defesa sanitária e inspeção.


#24 - Evaluation of skin prick test, exclusion diet and dietary challenge in the diagnosis of food allergy in dogs with chronic pruritus

Abstract in English:

Allergic dogs are commonly sensitized to food allergens. We evaluated the use of the prick test to diagnose food allergies in dogs with pruritus and the efficacy of an exclusion diet based on the test. The prick test was performed in 10 healthy dogs and 34 dogs with pruritus, of which 25 received an exclusion diet for 60 days and was challenged with positive food on the test. pVAS and CADESI-4 were assessed on days 0, 30, and 60 after re-exposure. As a result, two control group dogs reacted to a single food allergen, milk and wheat. Of the 25 dogs with pruritus that reacted to food allergens, 24 (96%) reacted to more than one food allergen, and only one (4%) reacted to a single food protein: pork. In the test group (n=25), there was a significant improvement of pVAS and CADESI-4 after 30 and 60 days of dietary exclusion, with significant worsening of the scores with food allergen challenge. In conclusion, the prick test can be used for screening food allergens to make an exclusion diet.

Cães alérgicos são comumente sensibilizados a alérgenos alimentares. Nós avaliamos o uso do teste de puntura para o diagnóstico de alergias alimentares em cães com prurido e a eficácia de uma dieta de exclusão baseada no teste. O teste de puntura foi realizado em 10 cães saudáveis e em 34 cães com prurido, dos quais 25 receberam uma dieta de exclusão por 60 dias e depois desafiados com o alimento que foi positivo no teste. pVAS e CADESI-4 foram realizados nos dias 0, 30, 60 e depois da reexposição. Como resultados, dois cães do grupo controle reagiram a um único alérgeno alimentar, leite e trigo. Dos 25 cães com prurido que reagiram aos alérgenos alimentares, 24 (96%) reagiram a mais de um alérgeno alimentar e apenas um (4%) reagiu a uma única proteína, que foi porco. No grupo teste (n=25), houve uma significativa melhora do pVAS e CADESI-4 depois de 30 e 60 dias de dieta de exclusão, com significante piora dos scores com o desafio alimentar. Como conclusão, o teste de puntura pode ser usado para selecionar alérgenos alimentares para fazer uma dieta de exclusão.


#25 - Fatal traumatic injuries in free-living wild Passeriformes and Psittaciformes birds in Central Brazil, 2006-2018

Abstract in English:

Traumatic injuries are a significant cause of death for birds worldwide, as they are at an increased risk of collisions and other injuries due to man-made environments. This study examined the frequency and morphological characteristics of fatal traumatic injuries in endemic and migratory Passeriformes and Psittaciformes from the Cerrado Biome, a biodiverse but threatened area in Brazil. Results showed that fatal traumatic injuries were found in 21.8% of birds (285/1305), mainly in spring and summer, during the birds’ reproductive period. The yellow-chevroned parakeet (Brotogeris chiriri) and Passeriformes from the Thraupidae family were the most affected. Nearly 70% of the fatal injuries observed were to the thoracic, pelvic limbs, and skull, and types of fractures and affected bones were thoroughly evaluated. Blunt traumas were one of the most frequent causes of injuries. Injuries affecting the appendicular skeleton and head represented significant causes of traumatic death for Passeriformes and Psittaciformes. The frequency of these fatal injuries has been increasing in recent years, which may be related to the remarkable environmental changes in the Cerrado Biome and jeopardize the survival of many bird species.

As lesões traumáticas são uma causa significativa de morte nas aves em todo o mundo, pois apresentam um risco maior de colisões e outras lesões devido aos ambientes degradados e criados pelo homem. Este estudo examinou a frequência e as características morfológicas das lesões traumáticas fatais em Passeriformes e Psittaciformes endêmicos e migratórios do Bioma Cerrado, uma área com rica biodiversidade, mas ameaçada no Brasil. Os resultados demostraram que as lesões traumáticas fatais foram observadas em 21,8% das aves (285/1305), principalmente na primavera e verão, durante a época reprodutiva das aves. O periquito-do-encontro-amarelo (Brotogeris chiriri) e Passeriformes da família Thraupidae foram as aves mais frequentemente acometidas. Por volta de 70% das lesões fatais observadas foram nos membros torácicos e pélvicos, e crânio, e os tipos de fraturas e ossos afetados foram minuciosamente avaliados. Os traumas contudentes foram as principais causas das lesões. As injúrias que afetaram o esqueleto apendicular e a cabeça representaram as mais importantes causas de morte traumática para Passeriformes e Psittaciformes. A frequência dessas lesões fatais vem aumentando nos últimos anos, o que pode estar relacionado às mudanças ambientais marcantes no Bioma Cerrado e colocar em risco a sobrevivência de muitas espécies de aves.


#26 - Urea poisoning in cattle: A brief review and diagnostic approach

Abstract in English:

Urea is an organic compound characterized as a white, solid, and hygroscopic substance. It is recognized as a source of non-protein nitrogen (NPN) and is widely used as a partial replacement for protein in cattle diets due to the ability of the ruminal microbiota to convert it into microbial protein. Despite the advantages of using urea, it also has limitations, particularly the proximity between metabolizable and toxic or fatal doses. Furthermore, for safe use, a period of adaptation is necessary for the animals. Poisoning is characterized by rapid and generally fatal development, which is frequent in non-adapted animals but can also occur in those with previous adaptations. The aim of this study was to characterize the clinical, epidemiological, and pathological aspects of urea poisoning through a brief review and a retrospective study. In addition, interviews were conducted with veterinarians who frequently send diagnostic material to the Laboratory of Anatomic Pathology of the “Faculdade de Medicina Veterinária e Zootecnia” (LAP-FAMEZ) to assess their perception of the outbreaks of urea poisoning. The objective was to obtain a comparative scenario between published cases and those received by the laboratory while considering the real situation of this condition in the field. During this retrospective study, only four outbreaks were investigated; in one, the diagnosis was possible through experimental reproduction. Of 35 interviewees, 88.9% said they had seen more than one case compatible with urea poisoning, but 87.5% did not perform a necropsy and/or send material to confirm the diagnosis. The results show that the reality of urea poisoning may be very distant from that reported in previous studies due to the difficulty often observed in the diagnostic approach, so we developed a flowchart aiming to provide a useful guide for field veterinarians.

A ureia é um composto orgânico, que se apresenta como uma substância branca, sólida e higroscópica, e é reconhecida como fonte de nitrogênio não proteico (NNP), sendo amplamente utilizada como fertilizante e também como substituto parcial de proteína em bovinos devido à capacidade da microbiota ruminal de convertê-la em proteína microbiana. Apesar das vantagens que envolvem o uso da ureia, ela também apresenta limitações, a principal delas é a proximidade entre doses metabolizáveis e tóxicas ou fatais, e para que seja utilizada com segurança é necessário um período de adaptação dos animais. A intoxicação é caracterizada por evolução rápida e geralmente fatal, sendo frequente em animais não adaptados, mas pode ocorrer naqueles com adaptação prévia. O objetivo deste estudo é caracterizar os aspectos clínicos, epidemiológicos e patológicos da intoxicação por ureia por meio de uma breve revisão e um estudo retrospectivo. Adicionalmente foram realizadas entrevistas com médicos veterinários que frequentemente enviam material para diagnóstico no Laboratório de Anatomia Patológica da Faculdade de Medicina Veterinária e Zootecnia (LAP-FAMEZ) com o objetivo de avaliar a percepção dos mesmos em relação aos surtos de intoxicação por ureia, na expectativa de se obter um quadro comparativo entre os casos recebidos pelo laboratório e levantamentos publicados e a real situação desta condição a campo. No período do estudo retrospectivo, foram acompanhados apenas quatro surtos, sendo que em um deles foi possível diagnosticar por reprodução experimental. Dos 35 entrevistados, 88,9% afirmaram ter atendido mais de um caso compatível com intoxicação por ureia, mas 87,5% deles não realizaram necropsia e/ou envio de material para confirmação do diagnóstico. Foi possível observar que a realidade da intoxicação por ureia pode estar muito distante daquela relatada em estudos anteriores, e devido à dificuldade muitas vezes observada na abordagem diagnóstica, desenvolvemos um fluxograma na expectativa de que ele constitua um guia útil para veterinários de campo.


#27 - Casuistry of xenarthrans treated at the Veterinary Hospital of the Federal University of Pará, Brazilian Amazon

Abstract in English:

The present study aimed to analyze the casuistry of xenarthrans treated at the veterinary hospital of the Federal University of Pará to quantify the most important cases for clinical care and identify the main species, sex, age group, origin, and destination of the animals admitted. The data were collected from January 2013 to August 2020. The variables analyzed were the number of animals by species, sex, age group, weight (kilograms), form of admission (rescue or guardianship), person responsible for the admission of the animal, origin, circumstances of admission, length of stay (days), and evolution or destination of the animal (release, captivity, death, euthanasia, or escape). Fisher’s exact test was used to determine whether disease incidence was related to sex and whether animal mortality was influenced by sex and disease. The chi-square test was used to determine whether mortality differed by age group. Ninety-two (92) xenarthrans were included: 66 Bradypus variegatus, 10 Choloepus didactylus, 15 Tamandua tetradactyla, and one Dasypus novemcinctus. The animals were mainly from the municipality of Castanhal (70%) and were acquired through rescue (88%, B. variegatus; 80%, C. didactylus; 100%, T. tetradactyla), a significant proportion of which was per natural persons (88%, B. variegatus; 80%, C. didactylus; 100%, T. tetradactyla). The most important circumstances that led to the admission of the animals were as follows: animals for examination (42%, n=39), young animals at risk (9%, n=8), hit by a vehicle (8%, n=7), respiratory disease (7%, n=6), fall from a tree (5%, n=5), and electrotrauma (5%, n=5). Release was the most common destination of animals after hospitalization (59% B. variegatus, 50% C. didactylus, and 66% T. tetradactyla). The sex of the animals did not affect the incidence of disease or mortality in any species. In contrast, mortality of B. variegatus was significantly higher in animals admitted with disease (p=0.000). Mortality in cubs of this species was higher than that in juveniles and adults (p=0.003).

O objetivo deste trabalho foi analisar a casuística dos Xenarthras atendidos no Hospital Veterinário da Universidade Federal do Pará (UFPA), a fim de quantificar as principais ocorrências para atendimento clínico, identificar as principais espécies, o sexo, a faixa etária, a procedência e destinação dos animais recebidos. Os dados foram obtidos entre janeiro de 2013 a agosto de 2020. As variáveis analisadas foram número de animais por espécie, sexo, faixa etária, peso (quilogramas), forma de recebimento (resgate, tutela), responsável pelo encaminhamento do animal ao hospital, procedência, circunstância de admissão, tempo de internação (dias) e evolução ou destinação do animal recebido (soltura, cativeiro, morte, eutanásia, fuga). O teste de Fisher foi utilizado para avaliar se a ocorrência de doenças estava relacionada ao sexo, bem como se a mortalidade animal foi influenciada pelo sexo e por enfermidades. O teste Qui-quadrado foi utilizado para avaliar se a mortalidade diferiu em relação à faixa etária. Um total de 92 animais Xenarthras foi admitido, composto por 66 Bradypus variegatus, 10 Choloepus didactylus, 15 Tamandua tetradactyla e um Dasypus novemcinctus. Os animais foram procedentes principalmente do município de Castanhal (70%), sendo o resgate a principal forma de aquisição dos animais (88%, B. Variegatus; 80%, C. Didactylus; 100%, T. tetradactyla), realizado em maior parte por pessoa física (88%, B. Variegatus; 80%, C. Didactylus; 100%, T. tetradactyla). O sexo dos animais não influenciou a ocorrência de doenças ou mortalidade em nenhuma espécie. Por outro lado, a mortalidade de B. variegatus foi significativamente maior naqueles animais admitidos com alguma enfermidade (p=0,000). A mortalidade em filhotes desta espécie foi maior quando comparada com as categorias jovem e adulta (p=0,003).


#28 - Detection and genotyping of Giardia duodenalis infecting pigs and small ruminants in the state of Piauí, northeastern Brazil

This study performed a molecular detection and characterization of Giardia duodenalis infecting pigs, goats and sheep in rural and peri-urban communities in the state of Piauí, northeastern Brazil, and proposed phylogenetic relationships among the characterized parasites. We assessed 52 fecal samples from pigs, 13 from goats, and 10 from sheep. A fragment of the β-giardin locus was PCR-amplified and sequenced. Overall, PCR-based G. duodenalis positivity was 11/52 (21.2%) in pigs, 2/13 (15.4%) in goats, and 2/10 (20%) in sheep. Seven out of 15 successfully amplified samples could be sequenced: three from pigs, two from goats, and two from sheep. Parasites from different hosts were found to belong to sub-assemblage AII. The phylogenetic analyses of the original G. duodenalis AII β-giardin sequences obtained from distinct host species and sequences of G. duodenalis recovered from humans available in GenBank suggest that the parasites are genetically related, supporting a local scenario of cross-host transmission.

Este estudo teve como objetivo detectar e caracterizar geneticamente amostras de Giardia duodenalis recuperadas de suínos, caprinos e ovinos em comunidades rurais do estado do Piauí, no nordeste do Brasil, propondo relações filogenéticas entre os parasitas caracterizados. Foram estudadas 52 amostras fecais de suínos, 13 de caprinos e 10 de ovinos. Uma região (560 pb) do lócus codificante da β-giardina foi amplificada por PCR e submetida a sequenciamento nucleotídico. A positividade para G. duodenalis pela PCR foi 11/52 (21,2%) em suínos, 2/13 (15,4%) em caprinos e 2/10 (20%) em ovinos. De 15 amostras amplificadas, sete puderam ser sequenciadas: três obtidas de suínos, dois de caprinos e dois de ovinos. Todas foram caracterizadas como pertencentes à subassemblage AII. Análises filogenéticas de amostras de G. duodenalis AII identificadas em diferentes hospedeiros, incluindo sequências de parasitas recuperadas de humanos e obtidas no GenBank, sugerem que os isolados têm alto grau de homologia. Os resultados apontam para um cenário de transmissão cruzada entre diferentes espécies de hospedeiros.


#29 - Detection of feline panleukopenia virus (Carnivore protoparvovirus 1) in free-ranging Panthera onca in Brazil

Abstract in English:

The decline in the jaguar population confirms how much the species is vulnerable to extinction in Brazil. It also indicates the degradation of its natural habitat’s environmental integrity and quality. Studies claim that large felids are susceptible to feline panleukopenia virus (FPV) and are presumptively diagnosed clinically in Brazil. A free-living jaguar (Panthera onca) cub was found unconscious and rescued due to a possible hit-and-run in the savannah of Mato Grosso. During recovery, it exhibited clinical and hematological signs consistent with FPV infection. The PCR was positive for FPV, with 99.61% identity between the FPV sequences available in the GenBank database through the BLAST tool. Due to habitat restrictions, certain diseases threaten wild cats and habitat encroachment by domestic animals can alter the pattern of spread of pathogens. We highlight the importance of the molecular diagnosis and phylogenetic analysis of FPV to elucidate how it has reached wild felids.

O declínio da população de onça-pintada confirma o quanto a espécie está vulnerável à extinção no Brasil, indicando também a degradação da integridade ambiental e da qualidade de seu habitat natural. Estudos afirmam que felinos de grande porte são suscetíveis ao vírus da panleucopenia felina (FPV) e são diagnosticados clinicamente de forma presuntiva no Brasil. Um filhote de onça-pintada (Panthera onca) de vida livre foi encontrado inconsciente e resgatado devido a um possível atropelamento no cerrado do Mato Grosso. Durante a recuperação, apresentou sinais clínicos e hematológicos compatíveis com infecção por FPV. A PCR foi positiva para FPV, com 99,61% de identidade entre as sequências de FPV disponíveis no banco de dados GenBank por meio da ferramenta BLAST. Devido a restrições de habitat, certas doenças ameaçam felinos selvagens e a invasão de habitat por animais domésticos pode alterar o padrão de propagação de patógenos. Destacamos a importância do diagnóstico molecular e da análise filogenética do FPV para elucidar como ele atinge os felídeos silvestres.


#30 - Parasitic macrofauna of Podocnemis unifilis from the Tocantins and Javaés Rivers, Eastern Amazon, Brazil

Abstract in English:

The parasitic fauna of Podocnemididae species is of great relevance to understanding the ecological relationships of the organisms in this genus. The parasites can have a symbiotic relationship with their hosts; however, environmental imbalances may change this relationship. Therefore, these parasites may become agents that generate zoonoses, especially in animals used by parasites as a food source. The aim of this study was to investigate the gastrointestinal parasitic fauna of specimens of Podocnemis unifilis. The animals were collected from the Tocantins and Javaés Rivers, both of which flow in the State of Tocantins, predominantly in the Cerrado territory. Samples of stomach and intestinal contents were fixed in 10% formalin buffered with sodium phosphate, transferred to 70% alcohol for analysis, and separated after 72 hours. Parasites were isolated and identified according to morphology and identification key. Four species of parasites were observed in 100% of specimens of P. unifilis, namely Nematophila grandis (Trematoda) and Paratractis hystrix, Paraorientatractis semiannulata, and Orientatractis leiperi (Nematoda). N. grandis was the only parasite that was not observed in the intestine; it appeared to adhere to the inner wall of the stomach. All nematodes were observed both in the stomach and intestine. These data expand the knowledge related to the P. unifilis parasitic fauna and the geographic distribution of these parasites in P. unifilis from the Tocantins and Javaés Rivers.

A fauna parasitológica das espécies de Podocnemididae é de grande relevância para o conhecimento das relações ecológicas desse grupo de vertebrados. Os parasitos podem ocorrer em simbiose com os seus hospedeiros sem causar danos, mas o desequilíbrio ambiental pode causar um desequilíbrio nessa relação e, assim, serem agentes geradores de zoonoses, especialmente em animais que são utilizados como recurso alimentar. Esta pesquisa fez o levantamento da fauna parasitária gastrointestinal de espécimes de Podocnemis unifilis. Os animais foram coletados no rio Tocantins e Javaés, ambos no Estado do Tocantins, em território predominante de Cerrado. As amostras de conteúdo estomacal e intestinal foram fixadas em formol a 10% tamponado em fosfato de sódio, transferidas para álcool 70% para análise e separação após 72 horas. Os parasitos foram separados e identificados de acordo com morfologia e chave de identificação. Foram encontradas quatro espécies de parasitos com prevalência em 100% dos espécimes de P. unifilis, sendo elas Nematophila grandis (Trematoda) e Paratractis hystrix, Paraorientatractis semiannulata e Orientatractis leiperi (Nematoda). Somente N. grandis não foi observado no intestino, sendo encontrado somente aderido à parede interna do estômago. Todos os nematoides foram observados no estômago e intestino. Esses dados ampliam o conhecimento sobre a fauna parasitária de P. unifilis, e aumentam o conhecimento sobre a distribuição geográfica desses parasitos em P. unifilis dos Rios Tocantins e Javaés.
